KPV: Research Chemistry and Literature
KPV is a tripeptide of lysine, proline and valine, written Lys-Pro-Val in three-letter code. The free acid has the formula C16H30N4O4 and an average mass near 342.4 daltons. The sequence is not arbitrary. It reproduces residues 11 to 13 of alpha-melanocyte-stimulating hormone, the C-terminal tripeptide of that thirteen-residue melanocortin, which is itself a proteolytic product of proopiomelanocortin.
Structure-activity work on the parent hormone narrowed a large part of the inflammatory-signalling literature onto this short C-terminal message, and the fragment entered the record under its single-letter contraction. KPV is therefore a minimal native fragment rather than a designed analogue. PubChem indexes the free tripeptide under the synonym MSH (11-13). Note that the sequence carries no aromatic residue. That detail matters later, at the detector.

Origin and identification
Alpha-melanocyte-stimulating hormone is a thirteen-residue melanocortin cleaved from proopiomelanocortin, and its chain terminates in Lys-Pro-Val. Through the 1980s and 1990s, work on which part of the parent peptide carried its inflammatory-signalling activity converged on that terminal triplet.
Getting, Schioth and Perretti made the comparison explicit in 2003, setting core and C-terminal fragments of the parent hormone side by side in a murine crystal-induced peritonitis model and in macrophage culture. Their dissection separated melanocortin-receptor-dependent activity attributable to the core sequence from activity retained by the tripeptide. Recessive yellow mice, which carry a non-functional melanocortin 1 receptor, were used to draw that line.
How the tripeptide is recognised has been answered less by receptor pharmacology than by transport biology. Dalmasso and colleagues reported in 2008 that KPV enters intestinal epithelial and immune cells through PepT1, the proton-coupled oligopeptide transporter that handles di- and tripeptides generally. That finding shifted the question from binding to uptake.
Preclinical research context
The cell-culture record is dominated by intestinal epithelial lines. Caco2-BBE and HT29-Cl.19A monolayers recur as the standard systems, usually alongside Jurkat T cells, with nuclear factor kappa B reporter activity and pro-inflammatory transcript levels as readouts. A separate strand ran in fetal rat alveolar type II epithelial cells. Haddad and co-workers tracked nuclear translocation of the p50, RelA and c-Rel subunits after endotoxin challenge and reported reversal of IkappaB-alpha phosphorylation and degradation by the Lys-D-Pro-Val analogue.
Rodent work sits almost entirely in colitis models. Dextran sulfate sodium and trinitrobenzene sulfonic acid colitis in C57BL/6 mice appear in the Dalmasso study. Kannengiesser and colleagues added the CD45RB-high T cell transfer model and repeated the dextran sulfate sodium protocol in animals lacking a functional melanocortin 1 receptor. Endpoints across this group of papers are histological colitis scoring, myeloperoxidase activity and mucosal cytokine profiles.
Outside the gut, the peritonitis model supplied a leukocyte-recruitment endpoint, counted as polymorphonuclear cell accumulation in the peritoneal cavity. A newer formulation literature has grown around the same tripeptide, using functionalised polymeric nanoparticles to concentrate it at inflamed colonic tissue in mice. Those papers are delivery-engineering studies and should be read as such, since the carrier rather than the peptide is normally the variable under test.
Analytical characterisation
Chromatographic purity is established by reverse-phase HPLC, conventionally on a C18 stationary phase with a water and acetonitrile gradient and trifluoroacetic acid as the ion-pairing modifier. KPV is short and polar, so it retains weakly and the gradient has to start shallow. Detection sits at low ultraviolet, 214 or 220 nanometres, where the amide bond absorbs. There is no tryptophan, tyrosine or phenylalanine in the sequence, so the 280 nanometre channel is blind to this peptide and a chromatogram recorded there proves nothing. Purity is reported as area percent of the integrated trace.
Identity is confirmed by mass spectrometry. Electrospray ionisation gives a singly protonated ion near m/z 343.2 against a monoisotopic neutral mass of 342.23. Intact mass alone is weaker evidence than it looks for a tripeptide of this composition, because Lys-Val-Pro and Pro-Val-Lys share the same formula and the same nominal mass. Tandem fragmentation resolves the b and y ion series and separates the intended sequence from those transpositions.
Alpine publishes lot-specific Certificates of Analysis issued by Janoshik Analytical, an independent testing laboratory. Each certificate carries the chromatogram with its integration table and the corresponding mass spectrum. Certificates are keyed to lot identifiers rather than to the catalogue entry, so a document describes the batch it was cut from and nothing beyond it.
Handling and storage
Lyophilised KPV is stable at ambient temperature across the span of a normal shipment, and transit warmth is not a failure condition for the dry solid. On receipt, move the vial to minus 20 degrees Celsius for long-term storage and keep it dark. Peptides degrade under light faster than most benches assume, and an amber vial or a closed box costs nothing.
Reconstituted material is a different proposition. In solution the peptide should be held at 2 to 8 degrees Celsius and the working window treated as short. Solution stability is governed by pH, buffer composition and any adventitious protease activity, none of which the dry powder has to contend with.
Repeated freeze-thaw is the most common avoidable source of loss. Split the reconstituted stock into single-use aliquots before the first freeze, label each with the lot identifier, and thaw only what a given experiment consumes. Record the reconstitution date on the vial. A stock of unknown age is not a reagent.
